ABSTRACT
Introduction We investigated the relationship between systematic regulators of inflammation and risk age-related macular degeneration (AMD), both wet dry forms, by using bidirectional, two-sample Mendelian randomization (MR). Methods performed bidirectional analysis genome-wide study (GWAS) data for 91 plasma proteins from 14,824 individuals European descent across 11 groups. Next, we utilized FinnGen consortium to AMD inverse- variance-weighted approach randomization. Additional analyses involved MR-Egger, Weighted median, mode, MR-PRESSO, MR- Steiger filtering techniques. Results identified 16 cytokines associated outcomes post FDR correction, higher levels fibroblast growth factor 19 leukemia inhibitory receptor were with decreased AMD, while tumour necrosis ligand superfamily member 14 increased AMD. Additionally, interleukin-10 subunit alpha signaling lymphocytic activation molecule Genetic susceptibility was elevated TNF-related activation-induced (TNFSF11), genetic TNFSF11, interleukin-18 1 (IL18R1), CUB domain-containing protein (CDCP1). Discussion This research enhances our understanding systemic in providing insights into etiology, diagnosis, treatment its forms.
