ABSTRACT
Podocalyxin (PODXL), a cell surface sialomucin expressed in diverse types of normal and malignant cells, mediates cellular adhesion to extracellular matrix cell-to-cell interaction. A previous study reported the expression PODXL protein on monocytes undergoing macrophage differentiation, yet this molecule other antigen presenting cells (APCs) its function immune system still remain undetermined. In study, we report that is human monocyte-derived immature dendritic at both mRNA levels. Following dendritric maturation using pro-inflammatory stimuli, level decreased substantially. Furthermore, found positively regulated by IL-4 through MEK/ERK JAK3/STAT6 signaling pathways. Our results revealed polarized distribution during interaction APCs with CD4 + T partially colocalizing F-actin. Notably, overexpression promoted their CD8 MHC-I, MHC-II, costimulatory CD86. addition, reduced translocation T-cell centrosome toward APC-contact site. These findings suggest regulatory role for responses, thus representing potential target therapeutic blockade infection cancer.
