ABSTRACT
M1-polarized macrophages can improve the body’s immune function. This study aimed to explore mechanism of Platycodon grandiflorus polysaccharide (PGPS t ) degrading SOCS1/2 protein through autophagy and promoting M1 polarization in 3D4/21 cells. Immunoprecipitation, confocal laser scanning microscopy, flow cytometry, intracellular co-localization were used detect expression related phenotypic proteins cytokines The results showed that PGPS significantly promoted mRNA IL-6, IL-12, TNF-α enhanced TNF-α, IL-1β, iNOS, CD80, CD86, indicating Next, effect degradation on cells was detected. downregulated level protein, but had no obvious SOCS1/2, degraded by activating lysosome system. Further research found under action 3-MA BafA1, upregulated LC3B II expression, which increased possibility LC3B, key component autophagy, bridging this connection SOCS1/2. interaction between LC3 identified indirect immunofluorescence Co-IP. percentage two after treatment, interacted with SOCS1 SOCS2. provides a theoretical basis for application treatment or improvement diseases macrophage regulating level.
