ABSTRACT
DNA damage repair (DDR) is critical in maintaining normal cellular function and genome integrity associated with cancer risk, progression, therapeutic response. However, there still a lack of thorough understanding the effects DDR genes’ expression level progression resistance. Therefore, we defined tumor-related score (TR-DDR score), utilizing levels 20 genes, to quantify tumor signature pathways tumors explore possible mechanism for among different cancers. The TR-DDR has remarkably predictive power tissues. It more accurate indicator response chemotherapy or immunotherapy combined tumor-infiltrating lymphocyte (TIL) G2M checkpoint than pre-existing predictors (CD8 PD-L1). This study points out that generally positive correlations patients advanced-stage, genome-instability, cell proliferation signature, while negative inflammatory response, apoptosis, p53 pathway signature. In context immune strongly positively correlates number T cells (CD4+ activated memory cells, CD8+ regs, Tfh) macrophages M1 polarization. addition, by difference analysis correlation analysis, COL2A1 , MAGEA4 FCRL4 ZIC1 are screened as potential modulating factors score. summary, light on new biomarker its guide strategies drug prediction.
