ABSTRACT
Introduction Tertiary lymphoid structures (TLSs) are analogues of secondary organs that contain various immune cells. The spatial distribution, maturation and composition TLSs have differential effects on prognosis, the roles in gastric adenocarcinoma (GA) not been revealed. Methods Thus, we evaluated prognostic value GA through analysis bulk RNA sequencing(RNA-seq) data from public databases validated our findings tumour samples Fudan University Shanghai Cancer Center (FUSCC) cohort. distribution,maturation, were analysed by reviewing H&E-stained sections multiplex immunofluorescence (mIF) staining. Results We found TLSs, especially with germinal centres (GCs) located invasive margin (IM), correlated prolonged overall survival (OS). Second, RNA-seq showed high dendritic cell (DC) scores a favourable factor patients for both GCs. In FUSCC cohort, DC-LAMP+ DCs weresignificantly enriched IM-TLSs GCs, suggesting potential correlation between activation milieu DC abundance.
