ABSTRACT
Intramuscular fat contributes to the improvement of meat quality goats. MicroRNAs (miRNAs) have been reported regulate adipocyte differentiation and maturation. The aim our study was clarify whether miR-10a-5p regulates goat intramuscular preadipocyte (GIPC) its direct downstream signaling pathway. GIPCs were isolated from longissimus dorsi, whose level measured at different time point induction. Adipogenic evaluated by Oil Red O BODIPY staining, expression changes adipogenic genes like ACC, ATGL, CEBPβ, PPARγ, etc. Related mechanisms verified qPCR, a bioinformatic analysis, dual-luciferase reporter assay, overexpression, siRNA transfection. staining both with gene detection showed that suppressed accumulation lipid droplets in inhibited differentiation. assay experiment revealed GIPC directly binding KLF8 3’UTR expression. Thus, results indicated inhibits targeting supply new target for deposition improvement.
