ABSTRACT
Aging is an important cause of cognitive dysfunction. Liuwei Dihuang decoction (LW), a commonly applied Chinese medicine formula, widely used for the treatment aging-related diseases in China. Previously, LW was confirmed to be effective prolonging life span and reducing oxidative stress aged mice. Unfortunately, underlying mechanism remains unclear. The aim this study interpret by which alleviates dysfunction related aging from perspective microbiota-gut-brain axis.All C57BL/6 mice (n = 60) were randomly divided into five groups: control, model, vitamin E (positive control group), low-dose high-dose groups 12 each group). Except those group, D-galactose subcutaneously injected other induce model. antiaging effect evaluated water maze test, electron microscopy, 16S rRNA sequencing, combined LC-MS GC-MS metabolomics, ELISA.Liuwei ameliorated hippocampal synaptic ultrastructure damage Moreover, decreased Proteobacteria abundance increased gut microbiota diversity Metabolomic analysis showed that associated with significantly differential 14 metabolites, mainly enriched apelin signaling, sphingolipid metabolism, glycerophospholipid metabolic pathways. Additionally, affected lipid metabolism Finally, we also found LW-regulated microbial species such as Fibrobacterota had potential relationships metabolites.In brief, improved function regulating through restoration homeostasis axis.
