ABSTRACT
Background Focal motor seizures that originate in the region are a considerable challenge because of high risk permanent deficits after resection. Deep brain stimulation subthalamic nucleus (STN-DBS) is potential treatment for epilepsy may enhance antiepileptic actions substantia nigra pars reticulata (SNr). Orexin and its receptors have relationship with both STN-DBS epilepsy. We aimed to investigate whether how STN inputs SNr regulate role orexin pathway this process. Methods A penicillin-induced epileptic model adult male C57BL/6 J mice was established evaluate efficacy modulating seizure activities. Optogenetic chemogenetic approaches were employed STN-SNr circuits. Selective receptor type 1 2 antagonists used inhibit pathway. Results First, we found high-frequency ipsilateral or bilateral effective reducing activity model. Second, inhibition excitatory neurons projections alleviates activities, whereas their activation amplifies In addition, circuits also reversed protective effect on Finally, observed reduced elevated expression during using combination selective activity. Conclusion helps reduce by inhibiting circuit. Additionally, show suppressing be promising therapeutic option future.
