ABSTRACT
Chronic, inflammatory pain is an international health concern that severely diminishes individuals' quality of life. Catalpol iridoid glycoside derived from the roots Rehmannia glutinosa possesses anti-inflammatory, antioxidant, and neuroprotective properties for treating multiple kinds disorders. Nevertheless, catalpol's impacts on its potential methods action are still unclear. The purpose this investigation to determine mechanism catalpol reduce behaviors in a rat model with complete Freund's adjuvant (CFA). Catwalk, Von-Frey, open field testing were performed behavioral assessment. Western blot analysis real-time quantitative PCR (RT-PCR) employed identify variations molecular expression, while immunofluorescence was utilized cellular localization. effectively reduced CFA-induced mechanical allodynia thermal hyperalgesia when injected intrathecally. Moreover, can regulate HDAC4/PPAR-γ-signaling pathway CFA spinal cord neurons. Meanwhile significantly decreased expression NF-κB/NLRP3 axis rats. In addition, both vivo vitro research revealed treatment inhibited astrocyte activation increase factor expression. Interestingly, we also found could alleviate peripheral by inhibiting tissue inflammation. Taken together, findings declared may inhibit rats targeting
