Zhilong Huoxue Tongyu Capsules' Effects on ischemic stroke: An assessment using fecal 16S rRNA gene sequencing and untargeted serum metabolomics

Zhilong Huoxue Tongyu capsule (ZHTC) is an effective traditional Chinese medicine compound for the treatment of ischemic stroke, which widely used in clinical stroke patients. However, it uncertain whether ZHTC affects through gut microbiota and serum metabolites. In this study, a rat model middle cerebral artery occlusion (MCAO) was prepared. By evaluating motor nerve function score, infarct size, brain tissue damage intestinal barrier damage, found that improved stroke-related symptoms MCAO rats. Using 16S rRNA gene sequencing, fecal microbial transplantation (FMT), untargeted metabolomics, spearman correlation analysis metabolites, we can regulate abundance p_Firmicutes, p_Bacteroidota,p_Proteobacteria, g_Prevotella, g_Lactobacillus, regulated 23 differential Spearman Arginine positively correlated with o_Clostridiales, c_Clostridia, negatively p_Bacteroidetes, c_Bacteroidia,o_Bacteroidales; L-Lysine f_Christensenellaceae; L-methionine o_Lactobacillales, f_Lactobacillaceae, g_Lactobacillus. Altogether, study shows first time ameliorate by modulating metabolic disturbances. This lays foundation further revealing causal relationship between ZHTC, dysbiosis, plasma metabolite levels provides scientific explanation ameliorating effect on stroke.
