ABSTRACT
Background: Calcium signaling is implicated in multiple processes including immune response that important tumor progression. Kidney renal clear cell carcinoma (KIRC) the most frequent histological type of with up to a third cases develop metastases. As result lack in-depth understanding mechanisms underlying KIRC, treatment options have been limited. Here, we aim comprehensively investigate landscape Ca 2+ channels, pumps and exchangers KIRC patients. Methods: The mRNA expression profiles gene variations 58 calcium-related genes (CRGs) patients normal control were downloaded from TCGA database. CRGs-related risk score was constructed quantify calcium patterns by using least absolute shrinkage selection operator (LASSO) regression. prognostic value, biological functions, therapeutic sensitivities based on then evaluated methods. Finally, key CRGs identified weighted co-expression network analysis (WGCNA). TCGA-CPTAC, GSE53757 datasets, as well human tissues used for validation. Results: had significant differences CRG expression, prognosis, functions between two clusters. determined. mutation burden, infiltration, checkpoints, targeted inhibitors remarkably different high low subtypes. high-risk subtype characterized immunosuppressive microenvironment poor prognosis. Meanwhile, several drugs showed distinct sensitivity TRPM3 protein significantly lower tumors than controls. Low associated prognosis Conclusion: Our study highlighted promising value tumors. evaluation will contribute predicting clinical therapy
