ABSTRACT
Background: Osteoarthritis (OA) is the most common joint disorder, lacking disease-modifying treatments. Adipose-derived mesenchymal stem cells (ADSCs) are adult multipotent stromal obtained from fat tissue, which holds great potential in treating OA. This study aimed to evaluate anti-OA efficacy of ADSCs preclinical and clinical facets explore underlying mechanism action. Methods:In vivo, a single dose 5 × 105 was injected into knee joints monoiodoacetate-induced OA rat model. The levels metabolic hypertrophic molecules (MMP13, Collagen II, X) chondrocytes were measured by immunohistochemistry. In vitro, cell viability assay conducted detect proliferation ability treated with conditioned medium (ADSCs-CM). Quantitative real-time polymerase chain reaction Western blot assays applied action ADSCs. Moreover, retrospective analysis determine safety on patients. Results: animal showed that significantly alleviated cartilage lesions rats, as confirmed downregulation MMP13 X upregulation II. vitro data ADSCs-CM promoted chondrocytes, restored IL-1β-induced abnormal expressions molecular markers IL-6, Aggrecan, MMP3, MMP13, X, ADAMTS5, ADAMTS9, SOX6, SOX9 chondrocytes. Such regulatory effects these anabolic, catabolic, suggested paracrine-based mode Furthermore, reduced pain repaired damage patients, no adverse events. Conclusion: demonstrated efficacy, safety, ADSCs, providing promising cell-based therapeutic option for treatment.
