Protective Effect of Danshen Zexie Decoction Against Non-Alcoholic Fatty Liver Disease Through Inhibition of ROS/NLRP3/IL-1β Pathway by Nrf2 Signaling Activation

ABSTRACT
Lipid metabolism disorders are a prominent characteristic in the pathological development of non-alcoholic fatty liver disease (NAFLD). Danshen zexie decoction (DZD) is Chinese herbal medicine that based on and has an effect regulating lipid mechanism. However, anti-NAFLD mechanism DZD remain unclear. In this study, we observed therapeutic NAFLD rats investigated its possible mechanisms. Sixty Sprague Dawley were randomly assigned to six groups: control group, model Yishanfu (polyene phosphatidylcholine) low, medium high-dose groups. High-fat diet (HFD) was fed establish model, each treatment group given corresponding drugs at same time for eight consecutive weeks. The results revealed obvious disorder injury induced by HFD alleviated with DZD, which verified decreased serum TC, TG, ALT, AST, FFA, as well alleviation hepatic steatosis. production ROS reduced after DZD. SOD activity GSH content increased treatment, while MDA level decreased. administration could decrease IL-1β IL-18 contents. Moreover, upregulated expressions Nrf2, HO-1, GCLC, GCLM, it suppressed NLRP3, caspase-1, GSDMD, GSDMD-N. conclusion, data showed can reduce accumulation, alleviate oxidative stress inflammation, inhibit pyroptosis rats, might be ascribed suppression ROS/NLRP3/IL-1β signaling pathway activation Nrf2. Overall, these indicated expected drug NAFLD.
