ABSTRACT
Background: Knee osteoarthritis (KOA), a chronic degenerative disease, is mainly characterized by destruction of articular cartilage and inflammatory reactions. At present, there lack economical effective clinical treatment. Zhuifeng Tougu (ZFTG) capsules have been clinically approved for treatment OA as they relieve joint pain manifestations. However, the mechanism ZFTG in KOA remains unknown. Purpose: This study aimed to investigate effect on TLR4/MyD88/NF-κB signaling pathway its therapeutic rabbits with KOA. Study design: In vivo, we established rabbit model using modified Videman method. vitro, treated chondrocytes IL-1β induce pro-inflammatory phenotype then intervened different concentrations ZFTG. Levels IL-1β, IL-6, TNF-α, IFN-γ were assessed histological observations ELISA data. The viability was detected Cell Counting Kit-8 flow cytometry. protein mRNA expressions TLR2, TLR4, MyD88, NF-κB Western blot RT-qPCR immunofluorescence observation p65 expression, respectively, inhibiting injury alleviating degeneration. Results: inhibited, levels blood cell significantly downregulated, consistent results. Both NF-κB, proteins that nucleus decreased groups. Moreover, promotes survival inhibits apoptosis chondrocytes. Conclusion: alleviates degeneration knee cartilage, chondrocytes, underlying may be inhibition TLR4/MyD88/NF-kB secretion factors.
