ABSTRACT
Introduction:Cynanchum auriculatum (CA) is usually used to treat digestive disorders, such as anorexia, enteritis, dysentery, and indigestion. Functional dyspepsia (FD) characterized by a group of symptoms associated with the gastroduodenal region. Recent pharmacological studies have demonstrated efficacy CA for treating FD. However, pharmacokinetics (PK) tissue distribution in physiological FD states still unclear. The present study aimed clarify differences PK parameters four major active components (baishouwu benzophenone, deacylmet-aplexigenin, qingyangshengenin, syringic acid) under both states. Methods: For this, normal rats were orally administered 10 mg/kg extract. Then, plasma (heart, liver, spleen, lung, kidney, brain, stomach, small intestine) samples obtained. rat tissues quantified developing validating fast reliable ultra-high-performance liquid chromatography-mass spectrometry method. Results: area concentration-time curve from time zero t (AUC0-t) baishouwu benzophenone was significantly lower than (p < 0.01). had 0.001) apparent volume clearance qing-yangshengenin higher 0.05) AUC0-t deacylmetaplexigenin qingyangshengenin. rapidly distributed into various tissues, main target organs activity stomach intestine. In addition, deacylmetaplexigenin, qingyangshengenin could cross blood-brain barrier, indicating that brain may be another organ treatment Discussion: These results indicate pathological state alters behavior characteristics acid extract, providing an experimental basis role treatment.
