Danggui Buxue decoction regulates the immune function and intestinal microbiota of cyclophosphamide induced immunosuppressed mice

ABSTRACT
Ethnopharmacological relevance Danggui Buxue decoction (DBD) is a traditional Chinese herbal formula. According to the theory of medicine, combination Astragali Radix (AR) and Angelica sinensis (AS) classic prescription tonifying qi enriching blood. DBD has functions hematopoietic, immune enhancement inflammation inhibition, usually used treat blood deficiency symptoms. Aim study Cyclophosphamide (CY) can inhibit humoral cellular immunity, leading overall disorder body, resulting in immunosuppressive (IS). Pre-laboratory studies confirmed immunomodulatory effects DBD, but its mechanisms have not been thoroughly studied. In this study, main purpose was determine on function intestinal mucosal barrier IS mice induced by CY, initially explored mechanism DBD. Materials methods 100 g AR 20 AS were accurately weighed 0.5 g/mL obtained boiling, filtration rotary evaporation. Then, group administered 5 g/kg gavage, positive 40 mg/kg levamisole hydrochloride, whereas those control model groups given corresponding volume normal saline gavage for 1 week. At end experiment, blood, spleen, thymus, ileum cecum contents all experimental collected aseptically. mouse intraperitoneal injection 80 CY three consecutive days. Pathomorphology observe physical intestine, flow cytometry detect splenic lymphocytes, immunohistochemistry content barrier-associated proteins, ELISA measure secretion ileal SIgA, qRT-PCR mRNA expression immune-related genes high-throughput sequencing analysis contents. Results alleviated spleen tissue damage restored impaired functions, such as increased thymus index CD4+/CD8+ subsets lymphocytes. addition, could increase villi length ratio crypt depth (V/C), decrease depth. Moreover, administration up-regulated ZO-1, Occludin, Claudin-1, MUC-2 ileum. And secretions sIgA ZO-1 also significantly improved. Furthermore, diversity gut microbiota, improve composition flora relative abundance beneficial genus, Bacteroides . Conclusion CY-induced decreasing T lymphocyte subsets. improves improving morphology up-regulating levels SIgA. regulates microbiota dysregulation increasing species richness, regulating phylum, class order Bacteroidetes
