ABSTRACT
Background The protective effect of estrogen on the vasculature cannot be explained only by its action through receptors ERα and ERβ. G protein-coupled (GPER)—which are widely distributed throughout cardiovascular system—may also involved in this response. However, little is known about GPER actions hypertension. Therefore, study we evaluated vascular response mediated using a specific agonist, G-1, spontaneously hypertensive rats (SHR). We hypothesized that G-1 would induce relaxing resistance mesenteric arteries from SHR both sexes. Methods concentration-response curves (1 nM-10 μM) were performed sexes (10–12-weeks-old, weighing 180–250 g). effects before after endothelial removal incubation for 30 min with inhibitors L-NAME (300 indomethacin (10 alone or combined clotrimazole (0.75 catalase (1,000 units/mL). immunolocalization was investigated, hydrogen peroxide (H 2 O ) ROS dichlorofluorescein (DCF) dihydroethidium (DHE) staining, respectively. Results activation promoted similar female male rats, but participation different mediators. Males appear to more dependent NO pathway, followed H females endothelium pathway. Conclusion These findings show agonist can way, albeit differential results contribute understanding essential
