Expression of Extracellular Vesicle PIWI-Interacting RNAs Throughout hiPSC-Cardiomyocyte Differentiation

ABSTRACT
Extracellular Vesicles (EV) play a critical role in the regulation of regenerative processes wounded tissues by mediating cell-to-cell communication. Multiple RNA species have been identified EV, although their function still lacks understanding. We previously characterized miRNA content EV secreted over hiPSC-cardiomyocyte differentiation and found distinct expression hiPSC-EV driving its vitro bioactivity. In this work, we investigated piRNA profiles derived from key stages hiPSC-CM maturation, i.e., hiPSC (hiPSC-EV), cardiac progenitors (CPC-EV), immature (CMi-EV), mature (CMm-EV) cardiomyocytes, demonstrating that EV-piRNA differs greatly identified. Only four were significantly deregulated one hiPSC-EV, three CPC-EV, as determined differential analysis on small RNA-seq data. Our results provide valuable source information for further studies aiming at defining bioactivity therapeutic potential EV.
