ABSTRACT
Purpose: This study assessed the biological reliability of peripheral human cytokines and adipokines, influence participant characteristics on total error. has essential application to interventional cytokine measurement ensure that reported results are interpreted with confidence. Methods: Participants (49% female, 18-85 years, n = 84) completed two consecutive-day testing sessions. provided a venous blood sample at same time day across consecutive days, under standardized presentation, including 24-h rested 12-h fasted conditions. Multiplex immunoassay was used assess inflammatory analytes from samples (predominantly plasma). Repeat measurements were conducted between-day for precision quantification, technical (technique) error negated provide an estimate (attributed presentation) Results: Whilst there no evidence statistically significant error, small amount consistently present most (∼3.3%/0.07 pg/ml), which largest leptin (7.3%/210 pg/ml). There also sex adiponectin (total model explained 6-7% variation), where females demonstrated greatest Conclusion: Biological in this should be applied any future or individual repeated concentrations over maintain best practice procedures (12-h fasted, rested). In cases, raw used, exceptions women adiponectin. approach will certainty improved reporting intervention efficacy.
