Histogen Layers Contributing to Adventitious Bud Formation Are Determined by their Cell Division Activities

ABSTRACT
Saintpaulia ionantha is propagated by adventitious buds in horticulture, and periclinal chimeric cultivars are usually difficult to propagate. However, some can be with buds, the mechanism of which has been unknown. Striped flower "Kaname", "Concord", "Monique" were used investigate what causes color separation shoot-derived plants tissue culture. These revealed have mutated flavonoid 3', 5' hydroxylase (SiF3'5'H), WDR1 (SiWDR1), or 3 (SiF3H), respectively, their L1 layer. From our previous study using all flowers from shoots colored pink, was epidermal mother plants' flowers. We "Concrd" we obtained not only monochromatic-colored same as plants, but also a monochromatic subepidermal color, striped plants. Histological observations that cells divided actively at 14 d after culture they involved formation cultured leaf segments "Kaname". On other hand, "Concord" "Monique", number subepidermis rather higher than cells, sometimes shoot formation. In addition, plant size L1-derived non-vigorous smaller those derived conclusion, into two types. One type high cell division activity layer, single flower-colored shoots. Another low several layers including cultivar capable propagation shoots, possibility shown L2 layer could form involving activity.
