ABSTRACT
Trichomes, which are classified as glandular or non-glandular, hair-like epidermal structures that present on aerial parts of most plant species. Glandular secretory trichomes (GSTs) have the capacity to secrete and store specialized metabolites, widely used natural pesticides, food additives, fragrance ingredients pharmaceuticals. Isolating individual is an essential way for identifying trichome-specific gene functions discovering novel metabolites. However, isolation difficult time-consuming. Here, we report a method isolate GSTs from leaf epidermis dispense with fixation using laser capture microdissection (LCM). In this study, 150 were captured efficiently Artemisia annua leaves enriched artemisinin measurement. UPLC analysis microdissected samples indicated specific accumulation secondary metabolites could be detected small number GSTs. addition, qRT-PCR revealed GST-specific structural genes involved in biosynthesis pathway highly expressed Taken together, developed efficient collect comparatively pure unfixed leaved, so relatively obtained intact. This can implemented metabolomics research purely cell populations has potential discover
