ABSTRACT
Paulownia tomentosa flower polysaccharide (PTFP) from dried cultured P. flowers, is widely known for its immunomodulatory activities. Here, PTFP was extracted using hot water extraction, followed by ethanol precipitation methods. Structural characterization of revealed scanning electron microscope, high-performance anion-exchange chromatography, gel ultraviolet and infrared spectral. Meanwhile, adjuvant action PTFT on the immune responses to classical swine fever vaccine in mice evaluated further proclaim regulatory effect PTFP. The results showed that a type heteropolysaccharide with dense, rough surface high molecular weight (667.02 kDa), mainly composed glucose (30.93%), rhamnose (29.99%), galactose (15.66%), arabinose (6.95%), mannose (5.52%), xylose (4.80%). chromatography suggested configuration may be spherical structure. spectrum confirmed functional groups chemical bond contained -OH, O-H, C-H, C=O, C-O, etc. Moreover, exhibited obvious enhancement improving concanavalin A (ConA), lipopolysaccharide (LPS), CSFV E2-stimulated splenocyte growth natural killer cell activity CSFV-immunized mice. Similarly, titers E2-specific IgG, IgG1, IgG2a, IgG2b antibodies IFN-γ IL-10 levels were distinctly increased treatment. Overall, macromolecular primarily containing rhamnose, possessed auxiliary vaccine.
