Transcriptomics yields valuable information regarding the response mechanisms of Chinese Min pigs infected with PEDV

ABSTRACT
Porcine epidemic diarrhea virus (PEDV) causes porcine (PED), a highly infectious disease, which has resulted in huge economic losses for the pig industry. To date, pathogenic and immune response mechanism was not particularly clear. The purpose of this study to investigate responses pigs infected with PEDV.In study, 12 Min were randomly selected without taking colostrum. At 3 days old, eight piglets 1 mL PEDV solution (10 TCID50/ml), remaining four handled by 0.9% normal saline. Within age 7 died considered as death group. Correspondingly, alive individuals classified into resistance Tissues duodenum, jejunum, ileum, colon, cecum, rectum three groups collected measure content. Additionally, jejunum used measurements analyses Hematoxylin-eosinstaining (HE), immunohistochemical sections, transcriptomics. phenotypes showed that viral copy numbers jejunal damage had significant differences between groups. We also observed transcriptome differentially expressed (DE) analysis 6,585 DE protein-coding genes (PCGs), 3,188 long non-coding RNAs (lncRNAs), 350 microRNAs (miRNAs), mainly involved metabolic pathways. Furthermore, specific molecules each group identified, 97 PCGs,108 lncRNAs, 51 miRNAs included ceRNA-regulated networks. By weighted gene co-expression network (WGCNA) transcription factor (TF) prediction, 27 modules 32 motifs ( E -value < 0.05) annotated 519 TFs detected. Of these TFs, 53 PCGs. In summary, promising key PCGs, related immunological detected provided new insights pathogenesis PEDV.
