ABSTRACT
Development of a post-transplant kidney transplant tolerance induction protocol involving novel total lymphoid irradiation (TLI) conditioning method in rhesus macaque model is described. We examined the feasibility acheiving to MHC 1-haplotype matched transplants by establishing mixed chimeric state with infusion donor hematopoietic cells (HC) using TomoTherapy TLI. The was hypothesized permit elimination all immunosuppressive (IS) medications while preserving allograft function long-term without development graft-versus-host-disease (GVHD) or rejection. An experimental group 11 renal recipients received and outcomes were compared control ( n = 7) that same but HC infusion. chimerism operational accomplished two group. Both withdrawn from IS continued maintain normal for 4 years rejection GVHD. None animals achieved when eliminated. This demonstrated inducing TLI non-human primate combined transplantation.
