ABSTRACT
In clinical practice, patients’ tissues are fixed and paraffin-embedded in order to enable histological diagnosis. Nowadays, those also used for molecular characterization. Formalin is the most fixative worldwide, Bouin’s solution some worldwide institutions. Among targets, micro RNAs (miRNAs), single-stranded non-coding comprised of 18 24 nucleotides, have been demonstrated be resistant fixation paraffin-embedding processes, with consequent possible application practice. present study, let-7e-5p, miR-423-3p, miR-92a-1-5p, miR-30d-5p, miR-155-5p, miR-200a-3p, miR-429 were investigated formalin matched solution-fixed high grade serous ovarian cancers by means real-time droplet digital PCR (ddPCR). Micro detectable analyzable both formalin- Bouin’s-fixed specimens, but on average, higher Ct values lower copies/µL found samples. Data from formalin-fixed samples correlated significantly targets ones, except let-7e-5p miR-155-5p. This study shows that miRNAs possibility, after proper data normalization, compare miRNA-based ones.
