ABSTRACT
Cisplatin-based chemotherapy causes intestinal mucositis, which patients immense suffering and hinders the process of cancer treatment. Dioscin is a natural steroid saponin that exhibits strong anti-inflammatory immunomodulatory properties. Herein, we investigate protective effect dioscin on cisplatin induced mucositis in rats from perspective gut microbiota barrier. We established rat model by tail vein injection cisplatin, concurrently treated with oral administration. Parameters, such as body weight, diarrheal incidence, D-Lactate levels, were assessed order to evaluate effects rats. Furthermore, biological samples collected for microscopic microbiota, integrity, immune inflammation analyses elucidate mechanisms mucositis. The results revealed administration significantly attenuated clinical manifestations, histological injury Besides this, markedly inhibited dysbiosis cisplatin. Meanwhile, partially alleviated junctions between ileum epithelial cells increased mucus secretion. Moreover, effectively TLR4-MyD88-NF-κB signal transduction pathway reduced secretion subsequent inflammatory mediators. These suggested cisplatin-induced part modulating microflora profile, maintaining integrity inhibiting response through pathway.
