ABSTRACT
Baculovirus infection can prevent the pupation of insects. Juvenile hormone (JH) plays a vital role in regulating insect molting and metamorphosis. However, molecular mechanism baculovirus preventing larvae by pathway is still unclear. In this study, we found that Mamestra brassicae multiple nucleopolyhedroviruses (MbMNPV) prolonged larval stage fourth instar Helicoverpa armigera (H. armigera) 0.52 d caused an increase JH titer. To identify genes contribute to H. armigera-MbMNPV interaction, analyzed mRNA expression profiles fat bodies infected MbMNPV. A total 3637 differentially expressed mRNAs (DE-mRNAs) were filtered out through RNA-seq analysis. These DE-mRNAs mainly enriched Spliceosome, Ribosome biogenesis eukaryotes, Aminoacyl-tRNA biosynthesis, Mismatch repair, RNA degradation signaling pathway, which are related virus infection. Real-time PCR was used verify sequencing results. find titer, all transcriptome JHE JHEH genes, down-regulated. MbMNPV-infected group significantly down-regulated compared with control RT-qPCR. We further proved degraded RNAi, ELISA, RT-qPCR bioassay, while hydrolysis almost be ignored. Knocking down HaJHE promoted receptor gene Met downstream Kr-h1, replication This study clarified larvae. The MbMNPV leads titer inhibiting JHE. promotes prevents armigera, replication. provides new insights into interaction mechanisms.
