[Clinical application value of combined detection of serum miR-378 and miR-21 in gastric cancer].

Objective: To investigate the clinical value of combined detection serum miR-378 and miR-21 in gastric cancer (GC). Methods: Eighty-seven patients with GC 78 colorectal cancer(CRC) from National Cancer Center/Cancer Hospital, Chinese Academy Medical Sciences were selected, 83 individuals undergoing healthy physical examination selected as controls. The levels detected by quantitative real-time PCR (RT-qPCR) (result data transformed log2 for analysis). Results: Relative expression -1.24, -3.25 -2.73 controls, CRC patients, respectively. Compared significantly decreased (both P<0.05). 0.11, 2.34 2.47 up-regulated Moreover, level was inversely associated tumor stage (P<0.05). However, showed no significant differences among different pathological characteristics (all P>0.05). area under receiver operating characteristic curve (AUC), sensitivity specificity miRNA-378 to diagnose 0.770, 82.0% 66.0%, respectively, 0.900, 85.0%, 88.0% miR-21, AUC, 0.930, 92.0% 87.0%, while AUC CEA CA-199 0.767, all four factors 0.946. Conclusion: have a certain effect on diagnosis GC.目的： 探讨血清miR-378和miR-21联合检测对胃癌的临床诊断价值。 方法： 选择中国医学科学院肿瘤医院经病理确诊的胃癌患者87例，结直肠癌患者78例，健康体检者83例，采用实时荧光定量PCR法检测血清样本中miR-378和miR-21的表达水平(以log(2)对结果进行转换)。 结果： miR-378在健康对照者、胃癌患者和结直肠癌患者血清中的相对表达水平分别为－1.24、－3.25和－2.73，miR-378在胃癌和结直肠癌患者血清中的相对表达水平明显低于健康对照者(均P<0.05)。miR-21在健康对照者、胃癌患者和结直肠癌患者血清中的相对表达水平分别为0.11、2.34和2.47，miR-21在胃癌和结直肠癌患者血清中的相对表达水平明显高于健康对照者(均P<0.05)。胃癌患者血清中miR-378的低表达与肿瘤的临床分期有关(P<0.05)，分期越晚，下降程度越大；而miR-21在胃癌患者血清中的升高程度与患者的临床病理特征无关(均P>0.05)。血清miR-378诊断胃癌的曲线下面积(AUC)为0.770，灵敏度为82.0%，特异度为66.0%；miR-21诊断胃癌的AUC为0.900，灵敏度为85.0%，特异度为88.0%；miR-378＋miR-21诊断胃癌的AUC为0.930，灵敏度为92.0%，特异度为87.0%。CEA＋CA-199诊断胃癌的AUC为0.767，CEA＋CA-199＋miR-378＋miR-21诊断胃癌的AUC为0.946。 结论： 联合检测血清中miR-378和miR-21的表达对胃癌具有一定的诊断效能。.
