ABSTRACT
Objective To investigate the effect of propofol on endothelial nitric oxide synthase (eNOS) and inducible (iNOS) expression in thoracic aorta hypertensive rats.Methods Healthy SD rats both sexes weighing 240-280 g were used this study.Hypertension was induced by subcutaneous deoxycorticosterone 25 mg/kg twice a week for consecutive 7 weeks.Sixty-four randomly divided into four groups (n = 16 each):hypertension group (group H),low,medium high dose ( P1,P2,P3 ).Groups P1,P2 P3 received infusion at rate 20,30 40 mg* kg- 1 · h- 3 h respectively,while H equal volume normal saline instead propofol.Mean arterial pressure (MAP) monitored recorded before,1 after start or infusion.All animals sacrificed intravenous administration.Blood samples collected taking out eyeballs determination serum NO concentrations nitrate reductase method.The eNOS mRNA,iNOS mBNA determined reverse transcription-polymerase chain reaction.The iNOS protein Western blot.Results Compared to H,MAP decreased significantly,the increased mRNA up-regulated,and down-regulated dose-dependent manner P < 0.05 0.01 ).Conclusion Propofol can down-regulate up-regudate cells promote release rats,Which is mechanism decreasing pressure.
