ABSTRACT
Objective
To observe the effects of Curcumin on cellular apoptosis rat retinal vascular endothelial cells (RRVEC) induced by high glucose.
Methods
Generation 4 cultured RRVEC were used in this experiment, and identified with anti-vWF factor antibody immunochemistry immunofluorescence. The divided into control group (5.5 mmol/L glucose), glucose (30 treatment glucose+30 μmol/L Curcumin), respectively. Flow cytometry was to measure reactive oxygen species (ROS) level apoptosis. expression intensity location nuclear (NF)-κB p65 three groups detected immunochemistory. Bcl-2 Bax protein Western blot test.
Results
Immunostaining showed that positive for vWF factor. flow ROS higher than (t=8.677, P=0.000), but less (t=40.957, P=0.000). Compared group, decreased significantly (t=6.568, different among (F=325.137, (t=12.818, Immunochemistry NF-κB expressed strongly nuclei cytoplasm significant differences (t=8.322, results demonstrated compared Bcl-2/Bax ratio (t=4.362, 6.449; P=0.005, 0.001) increased (t=3.813, P=0.009) statistically differences. (t=2.577, 3.059; P=0.042, 0.022) (t=3.831, group.
Conclusion
Curcumin could suppress glucose. mechanism protecting may be via regulating signal pathway.
