ABSTRACT
To investigate the influence of PI3K/AKT signaling pathway on proportion and characteristics stem-like CD90(+) subpopulation human hepatocellular carcinoma (HCC) cell line MHCC-97.MHCC-97H cultures were treated with inhibitor LY294002. The was determined by flow cytometry, expression related proteins measured Western blot. clonogenicity CD90(-) cells plate colony formation assay. tumorigenicity compared between subpopulations (with different concentrations) in xenograft experiments nude mice, changes after addition LY294002 evaluated. CD90, SHP2, P-AKT, AKT xenografts examined. Data analyzed using t test.LY294002 capable reducing HCC stem from 2.98%±0.08% to 0.78%±0.08% (t = 32.400, P < 0.01) 95.13%±3.78% 61.82%±7.23% 7.617, 0.01). However, it showed no significant effect subpopulation. also reduced P-AKT cells, but did not significantly affect AKT. had inhibitory cells.The MHCC-97H has is dependent pathway.
