Effect of metformin in vivo on the inhibition of inflammation and joint protection of the collagen-induced arthritis rat model

Objective To verify the effect of metformin in vivo on inhibition inflammation and joint protection collagen-induced arthritis (CIA) rat model.Methods Under 4 ℃,dissolved type]Ⅱ collagen into acetic acid.In order to made mixture emulsion,blended it with complete freund's adjuvant 1∶ 1.Gave several spots intraperitoneal injection 0.3 mL emulsion tail skin around anus.Repeat experi-mental operation 7th day strengthen induction.After success induction,gave CIA models high doses metformin(100 mg.kg-1 · d-1),low metformin(50 mg kg-1 d-1) methotrexate (2.7 w-1) for treatment by gavage day.Evaluated score each group rats measured width talus every two days.Executed drew materials 28th day,then shot X-ray films ankle detected levels inflammatory anti-inflammatory factors serum Elisa.Results After continuous observation foot scoring measurement from width:arthritis was obviously lower than other induced groups (P ＜ 0.05),talus is also much smaller(P 0.05) ; rest has a certain reducing compared 0.05).X-ray radiography show:high can significantly save structure make clearance clear; are not better dose group,also severe soft tissue swelling,bone absorption,joint disintegration damage.Elisa show:High level factors-TNF-α,IL1 β,IL-6 0.05),also rise factor-IL-10 obviously(P 0.05).Low MTX have similar 0.05),but more significant metformin.Conclusions Metformin Inhibit protect model.
