ABSTRACT
Objective
To develop detection methods for porcine circovirus type 1 (PCV1) — the adventitious agent in rotavirus vaccine contamination incident, African green monkey kidney cell (Vero cell).
Methods
Six days after PCV1 infection, virus replication and proliferation Vero cells were tested with qualitative PCR, quantitative PCR (qPCR), electron microscopy (EM) situ hybridization (ISH).
Results
PCR products amplified from PCV1-infected DNA extracts presented specific band agarose gel electrophoresis. According to qPCR, copy number increased 794.3 times of original level. The limits qPCR both 102.0 copies/ml. Characteristic viral particles observed by EM. ISH showed dark positive signal infected cells.
