Research on the difference of C/EBP-α gene on hepatocyte and hepatic stellate cells in the autophagy and apoptosis

ABSTRACT
Objective To investigate the difference of autophagy and apoptosis hepatocyte hepatic stellate cells inducted by CCAAT enhancer binding protein -α(C/EBP-α) vitro study. Methods The adenovirus mediated C/EBP-α gene (Ad-C/EBP-α) were constructed, BRL-3A cell lines HSC-T6 infected with 50, 100, 200 MOI, situation was detected flow cytometry. The expression marker LC3Ⅰ/LC3Ⅱ application Western blot method. Then, differences in pathways 2 induced investigated. Results Flow cytometry showed that line significant 50 MOI line, dose increased, rate further upward trend (P<0.05). There no change line. HSC-T6LC3Ⅰ/LC3Ⅱ from level significantly decreased, higher, decline greater, downward line. Conclusions The has a dose-dependent inhibitory effect on cells, which apoptosis, but it hepatocytes.
