Effects of epigallocatechin-3-gallate on expression of glial fibrillary acidic protein in optic nerve crush model

ABSTRACT
Background Our previous study demonstrated that epigallocateehin-gallate(EGCG),an active ingredient of green tea,has protective effect on optical nerve after optic crush.Astrocyte was proved to play key role in the repair tissue,but influence EGCG astrocyte is unclear.Glial flbrillary acidic protein (GFAP) a special marker for astrocyte. Objective The aim this investigate expression GFAP tissue crush. Methods Seventy-two clean Wistar rats were randomly divided into normal control group,sham+EGCG group,optic crush+normal saline group(vehicle group),optic crush+EGCG group.Optic crush models established by clamping 60 seconds minitype clipper with force 40 gram.Only ocular cut sham group.Normal solution or EGCG(25 mg/kg)was intraperitoneally injected daily 5 days consecutively and orally administered(2 mg/kg)daily afterwards.The detected immunohistochemistry quantified Western blotting analysis day 7,14 28 modeling. Results lmmunochemistry showed weakly expressed both group sham+EGCG sliSht brown staining tissue.The deeply seen vehicle group,and intensity weakened compared modeling.Western revealed level rat significantly enhanced comparison group(P<0.01).On 7 14 modeling,the levels evidently decreased group(P<0.05).However,on modeling,no significant difference wag found between erush+EGCG group(P>0.05). Conclusion down-regulates crush-induced therefore attenuates activity astrocytes,suggesting might reduce formation glial scar.
