ABSTRACT
Background
Stem cell transplantation represents a promising treatment option for patients suffering from degenerative disorders. Accumulating evidences indicate that mesenchymal stem cells (MSCs) are able to differentiate into retinal pigment epithelial (RPE) -like cells. However, MSCs difficult obtain. Human adipose (ADSCs) proved have similar properties MSCs, but relevant study is less.
Objective
This was assess the feasibility of human ADSCs differentiating RPE-like and safety its application in vivo.
Methods
The third generation were incubated 6-well plate, 100 ng/ml growth factor, 50 μmol/L taurine 5×10-7 mol/L retinoic acid added medium 12 hours after cultured induce cells, conventional used as control group. Induced traced with PKH26, Pan-cytoke ratin (Pan-CK) monoclonal antibody identify under fluorescence microscope. suspension 1 μl intravetreally injected right eyes 6 BALB/c mice, equal volume PBS same way another mice. The animals sacrificed month injection, morphology examined by histopathology optical ultrastructure ganglion (RGCs) transmission electron use care complied Regulations Administration Affair Concerning Experimental Animals State Science Technology Commission.
Results
Cultured grew well slender polygone shape. Cell membranes showed red PKH26 induced. In addition, Pan-CK expressed induced response absent One intravitreal located on surface, clear No abnormality RGCs seen microscope.
Conclusions
Human can induction. mark well. There no adverse effect retina injection short-term duration mice.
