ABSTRACT
Studies on the relationship between obesity and bone have recently become widespread. The aim of this study was to investigate effect bone, utilizing a diet-induced obese mouse model, explore role free fatty acids (FFAs) in osteogenesis/adipogenesis marrow-derived mesenchymal stem cells (BMSCs). An model established by high-fat diet (HFD). Proximal femurs were collected at sacrifice, mineral density (BMD) proximal measured dual-energy X-ray absorptiometry. Bone histomorphometry performed using undecalcified sections femurs. differentiation BMSCs assessed colony formation assays gene expression analysis. In vitro, various osteogenic adipogenic genes determined real-time quantitative PCR after exposure conditioned medium (CM) from FFA-treated 3T3-L1 adipocytes. Western blotting further analyze representative protein PPARγ Runx2. BMD trabecular thickness significantly greater HFD mice than control mice. CFU-osteo assay showed increased osteogenesis BMSCs. mRNA level Runx2 higher, while Pref-1 lower compared BMSCs, Sox9 up-regulated CM adipocytes, CEBP-α down-regulated. Osteogenesis increased, adipogenesis decreased. conclusion, HFD-induced may play protective concomitantly promoting suppressing through factors secreted
