ABSTRACT
Metabarcoding is invaluable for understanding trophic interactions, enabling high-resolution and rapid dietary assessments. However, it requires a robust DNA barcode reference library accurate taxa identification. This dataset has been generated in the framework of InBIO Barcoding Initiative (IBI) Agrivole project. The integration these two projects was crucial, as aimed to investigate niche small mammals Trás-os-Montes Region through metabarcoding, which required reliable plant this same region. Given large number species not yet represented international databases, survey local plants essential fill gap. Thus, study created an database Portugal. current contains 632 vascular samples, all morphologically identified belonging 435 species. represents 14% 38.7% total known Portugal area, respectively.Of 1781 sequences provided dataset, 1099 contain new information (61.7%) at different levels: 254 (13.6%, ITS2: 41, trnL-ef: 126, trnL-gh: 87) are completely GenBank and/or BOLD databases time publication, 438 (24.6%, 59, 173, 206) records given 407 (22.9%, 187, 206, 14) provide additional (e.g. bp length, intraspecific genetic variability); remaining 682 (38.3%) equal (100% identity) already publicly available Overall, significant contribution knowledge public libraries. one releases IBI database, provides distributional data several taxa. All vouchers deposited Herbarium Museum Natural History Science University Porto (MHNC-UP) their barcodes Barcode Life Data System (BOLD), NCBI online International Nucleotide Sequence Database Collaboration (INSDC).
