ABSTRACT
The number, type, and location of cytokine- Ab-secreting cells activated in mice immunized boosted with plasmid DNA encoding the circumsporozoite protein malarial parasite Plasmodium yoelii (PyCSP) were monitored. initial humoral response was localized to draining lymph nodes characterized by production IgG1 anti-PyCSP Abs Th2 cytokine IL-4. In contrast, secondary dominated IFN-gamma (a Th1 cytokine) secretion IgG2a spleen. PyCSP mRNA detected only quadriceps muscles (sites injection), yet these sites lacked either or cells. These findings indicate that circulating lymphocytes encounter plasmid-encoded Ag muscle bed, initiate a nodes, then seed distal lymphoid organs. Profound differences observed between primary immune responses induced immunization, which may influence vaccine efficacy.
