Objective: To evaluate improving rheology anti-inflammatory properties C. extract (CO) its active fraction (ACO) on acute model Wistar rats characterize correlative constituents. Materials Methods: CO at 0.26, 0.53, 1.04 g/kg ACO 0.38, 0.75, 1.5 were administered to 3 days. Whole viscosity, plasma levels interleukin-6 (IL-6), nitric oxide (NO), tumor necrosis factor alpha (TNF-α), cyclooxygenase-2 (COX-2) measured. HPLC-QTOF/MS/MS method was identify major constituents ACO; two representative components (cyasterone chikusetsusaponin IV) from thrombin-induced human umbilical vein endothelial cells damage also assessed by thromboxane A2 (TXA2), endothelin (ET), malondialdehyde (MDA), COX-2, synthase (eNOS), superoxide dismutase (SOD). Results: significantly reduced whole IL-6, NO, TNF-α, COX-2 vivo. Forty compounds identified ACO, mainly as phytoecdysteroids saponins. Cyasterone IV could inhibit TXA2, ET, MDA, promote activities eNOS SOD vitro. Conclusion: possessed significant effects showed anti-inflammatory, antioxidant, anticoagulant Keywords:Acute rats, Kuan, HPLC-QTOF/MS/MS, rheologyView:PDF (2.74 MB) Full Text
