ROCK inhibitors alleviate myofibroblast transdifferentiation and vascular remodeling via decreasing TGFβ1-mediated RhoGDI expression

ABSTRACT
The aim of this study was to investigate the effects Rho GDP dissociation inhibitor (RhoGDI) on TGFβ1-mediated vascular adventitia myofibroblast transdifferentiation and inhibition ROCK inhibitors. Myofibroblast remodeling model were induced by TGFβ1 in vitro balloon injury vivo. H&E (Hematoxylin & Eosin) PSR (Picrosirius Red) staining used observe morphology while immunofluorescence, immunohistochemistry, Western blotting measure protein expression. Fasudil treatment reduced expression TGFβ1, RhoGDI1, RhoGDI2 addition rat model. α-SMA, TGFβRI, phospho-TGFβRI, RhoGDI2, collagen secretion human aortic adventitial fibroblasts (HAAFs). These diminished after with Y27632. Suppressing both RhoGDI1 also blocked TGFβ1-induced α-SMA HAAFs. Moreover, TGFβR RhoGDI2. data suggested that inhibitors alleviate decreasing RhoGDI.
