Insufficient ability of Rac1b to perturb cystogenesis

ABSTRACT
Rac1b is frequently expressed in a number of human cancer cells. It still unclear, however, whether causes morphological abnormalities epithelial tissues. To investigate induces changes 3-dimensional structures, we utilized an auxin-dependent protein expression system, which enabled us to rapidly induce and evaluate function MDCK (Madin-Darby Canine Kidney) cysts, model for polarized structure. Cells carrying the wild-type Rac1, constitutively active Rac1V12 gene were morphologically indistinguishable from normal, when their coding proteins not expressed. However, upon induction, Rac1V12, but Rac1 or Rac1b, significantly induced luminal cell accumulation. Live imaging with cycle indicators showed that promoted progression. From these results, concluded per se cannot proliferation. Rather, it considered may participate progression malignancy.
