ABSTRACT
The aim of this study is to know whether silibinin has an anticancer effect on triple negative breast cancer xenograft model using MDA-MB-468 cells. To establish the model, we injected cells into female Balb/c-nude mice. After establishing a oral was administered tested mice in way 200 mg/kg for 45 days. difference mean tumor volume between fed and control analyzed. epidermal growth factor receptor (EGFR) phosphorylation analyzed by Western blotting. expression VEGF, COX-2, MMP-9 genes tissue real-time polymerase chain reaction (PCR).In cells, found that administration significantly suppressed (silibinin treated vs. mice; 230.3 ± 61.6 mm(3) 435.7 93.5 mm(3), P < 0.001). EGFR inhibited treatment with 50 µg/mL silibinin. In real time-PCR analysis obtained from sacrificed mice, gene MMP-9, COX-2 51.8%-80% smaller group than can also verify similar result blotting analysis. We verified had preventing eventually production expression. Finally, models decreased after Silibinin.
