The caspase-3 inhibitor (peptide Z-DEVD-FMK) affects the survival and function of platelets in platelet concentrate during storage

ABSTRACT
Background Although apoptosis occurs in nucleated cells, studies show that this event also some anucleated cells such as platelets. During storage of platelets, the viability platelets decreased, lesions were observed, and underwent apoptosis. We investigated effects caspase-3 inhibitor on survival function after different periods storage. Methods Platelet concentrates obtained from Iranian Blood Transfusion Organization plastic blood bags. Caspase-3 (Z-DEVD-FMK) was added to These bags along with control which no stored a shaking incubator at 22℃ for 7 days. The Z-DEVD-FMK functionality analyzed by assessing their ability bind von Willebrand factor (vWF) aggregate presence arachidonic acid ristocetin. Cell surveyed MTT assay. Results At day 4 storage, ristocetin-induced platelet aggregation significantly higher inhibitor-treated (test) than samples; difference not significant 7. There acid-induced between test samples. However, binding vWF assay revealed samples both days study. Conclusion Treatment could increase survival.
