ABSTRACT
In this study, we aimed to evaluate the antioxidant, anti-apoptotic, osteoblastic and hypolipidemic effects of thymoquinone (TQ) treatment on steroid-induced osteonecrosis femoral head (ONFH) model in rats.A total 24 rats were randomly divided into four groups: control group administered saline; TQ 10 mg/kg/day orally; lipopolysaccharide/methylprednisolone (LPS/MPS) 20 µg/kg intraperitoneally LPS 40 mg/kg intramuscularly MPS establish ONFH model; LPS/ MPS+TQ both LPS/MPS and, then, once daily for weeks. All sacrificed after intracardiac blood collection their right femurs removed.Micro-computed tomography showed a higher bone mineral density lower porosity, Tr. Sp Sep data detected LPS/MPS+TQ group. histopathology, increased significantly decreased compared (p=0.0077). Histomorphometric examination revealed that percentage BV/TV was other groups (p<0.01 p<0.05, respectively), while it reached normal rates Immunohistochemically, angiogenesis indicators (8-hydroxy-20- deoxyguanosine [8-OHdG], malondialdehyde [MDA], B-cell lymphoma [Bcl-2], caspase-3, vascular endothelial growth factor [VEGF]) improved tissue serum with TQ. Furthermore, reduced low- high-density lipoprotein cholesterol ratio carboxy-terminal type 1 collagen crosslink (CTX) serum.Vascular hematopoietic cell damages occur due deoxyribonucleic acid (DNA) oxidative lipid peroxidative an can be successfully ameliorated by administration. This antioxidant anti-apoptotic may promising option early stage osteonecrosis.
