ABSTRACT
It is essential to analyze rare mutations in many fields of biomedical research. However, the detection usually failed due interference predominant wild-type DNA surrounded. Herein we describe a sensitive and facile method detecting point mutation on basis allele-specific amplification emulsion PCR. The identification selective are accomplished one-pot reaction. primers coupled magnetic beads allow exclusive enrichment mutant amplicons. productive bearing amplicons subsequently stained with fluorescent dyes. Thus, percentage as low 0.1%, can be detected by analysis. relative percentages among different samples roughly accessed counting fraction positive through flow cytometry.
