ABSTRACT
Transgenic potato lines of cv. Desiree containing the DREB1A gene driven by rd29A promoter were generated using Agrobacterium-mediated transformation. The morphological appearance 120 transgenic was classified into three categories as determined in vitro test-tube evaluation. Southern blot analyses genomic DNA conducted restriction enzymes HindIII and DraI. There significant differences between DSC quantitative salinity-tolerance evaluations at 1 M NaCl. Two recognized highly tolerant to salinity based on Duncan multiple range testing. Furthermore, there a correlation mean tolerance level copy number estimated from hybridization experiments. Northern experiments subsequently done cDNA probe with different levels tolerance. Salt-tolerant expressed substantially more transgene 2 5 h salt treatment, after which expression returned basal levels. These observations suggest that transfer rd29A::DREB1A can be used increase important agricultural crops, such tetrasomic polyploid potatoes, occurs diploid model species, Arabidopsis.
