Rutin Inhibits Hepatic and Pancreatic Cancer Cell Proliferation by Inhibiting CYP3A4 and GST

ABSTRACT
Abstract: Background: Digestive cancer is among the major causes of mortality and morbidity worldwide. Rutin, a bioactive secondary metabolite belonging to flavonoids distributed in many fruits vegetables has shown anti-proliferative, anti-cancer, neuroprotective activities. In this study, antiproliferative, antioxidant apoptotic activities rutin on hepatic pancreatic cell lines were investigated. Materials Methods: The effect cellular viability was monitored by SRB assay. Increasing activity caspases (3/7) used as an indicator apoptosis. Additionally, anti-inflammatory evaluated after measuring amount prostaglandin E2 (PGE2) produced through DPPH free radical scavenging assays, respectively. Moreover, inhibitory both CYP3A4 GST enzymes also been evaluated. Results: According data presented here, anti-proliferative raises number 3/7 investigated lines. Conclusion: HepG-2 cells showed highest growth inhibition, followed BxPC-3, Huh-7, MiaPaCa-2, Suit-2, normal line HPDE. Rutin inhibited cyctochrome P450 enzyme (CYP450 3A4) glutatihione-S-transferase activity, with dose-dependent inhibition. Furthermore, suppression PGE2 synthesis BxPC-3 supported rutin's action (high COX-2 expression). Keywords: Pancreatic Liver cancer, Apoptosis Anti-inflammatory, Antioxidant.
