Investigation on Toxoplasma gondii by Polymerase Chain Reaction and Loop-Mediated Isothermal Amplification in Water Samples from Giresun, Turkey

ABSTRACT
Toxoplasmosis is generally asymptomatic in immunocompetent subjects, however serious manifestations of the disease may develop immunocompromised patients and pregnant women. The mean Toxoplasma gondii seroprevalence which approximately 40% Turkish population, indicates high risk for development acute toxoplasmosis those cases. One transmission ways T.gondii consumption contaminated water. aim this study was to detect presence environmental drinking water samples by using standard polymerase chain reaction (PCR) loop-mediated isothermal amplification (LAMP) methods. A total 96 samples, them 76 were collected from creeks Giresun province center its various districts (Piraziz, Bulancak, Keşap, Espiye) 20 included study. precipitated aluminium sulfate method DNAs isolated pellets formed with sucrose gradient method. PCR LAMP applied DNAs, use primers F3 B3 specific B1 gene parasite. In our study, detected none LAMP, positive 13.2% (10/76) samples. Both methods yielded same results stations that Aksu creek center; Gelivera Espiye; Yolağzı, Keşap Login Bridge Keşap; Karadere İncivez Bulancak; Piraziz Çayırağzı counties.
