ABSTRACT
This study aimed at the generation of a stable transformed silkworm BmN cell line which can continuously express human interferon-λ2 (IFN-λ2) gene, and investigated antiproliferative activity this recombinant IFN-λ2. Silkworm cells were transfected with vector pIZT/V5-His harboring IFN-λ2 gene. After pIZT/V5- His- hIFN-λ2 vector, stably expressing gene selected using Zeocin. Following two months screening, was obtained. Stable be maintained lower Zeocin concentration. The representing 26 kDa protein band detected by SDS-PAGE Western blotting. expression level hIFN- λ2, determined ELISA, about 8.142 ng in 4 × 10 5 cells. MTT assay. 50% inhibitory concentrations (IC 50 ) on A549 (lung cancer cells), HL60 (acute promyelocytic leukemia BEL-7402 (liver cells) M231 (breast approximately 3.21, 2.84, 6.29 9.32 ng/ml, respectively. In summary, Human expressed line, demonstrated to tumor vitro . Key words : protein, expression, activity.
